DMRIE exhibits good stability under physiological conditions. The presence of hydrophobic lipid tails allows it to form stable liposomes or lipoplexes, protecting the encapsulated nucleic acids from degradation. Firstly, DMRIE has a positively charged quaternary ammonium headgroup, which enables it to interact with negatively charged nucleic acids, such as DNA or RNA. This electrostatic interaction promotes the formation of stable complexes, called lipoplexes, between the cationic lipid and the genetic material.

 

Secondly, DMRIE has a hydrophobic tail composed of two myristoyl chains. This hydrophobic region allows the cationic lipid to insert into and disrupt the lipid bilayer of cell membranes, facilitating the delivery of the nucleic acids into target cells. The hydrophobic tail also contributes to the stability of the lipoplexes by providing a structural support.
